Quantification of 4EBP1 phosphorylation with PICO

In this application note we detected and quantified 4EBP1 and its phosphorylated form using PICO technology. Using Absolute Quantification (AQ), we could show that upon treatment of BT474 or MCF7 cells with the kinase inhibitor Dactolisib, the phosphorylation of 4EBP1 is decreased ~200 fold. 

Therefore, PICO is a powerful technology to quantify post-translational modifications and to monitor the activity of signaling pathways. 

 

Quantification of extracellular vesicles (EVs) with PICO

In this application note we detected and quantified extracellular vesicles (EVs) from serum or cell supernatant using PICO technology. The EVs were measured with two different detection strategies: 1) using an anti-CD63 antibody, halved and labeled with two different PICO Labels, we quantified CD63+ EVs, while 2) using both anti-CD63 and anti-CD9 antibodies, we quantified CD63/CD9+ EVs. These findings highlight the versatility of PICO in sensitive and accurate detection of various protein targets.

PICO vs. Western Blot

In this application note we compared the sensitivity of PICO to western blot. We chose to detect the breast cancer biomarker HER2/ErbB2 in two different breast cancer cell lines, BT474 and MCF7. Through our analysis, we found that PICO demonstrated approximately 200-fold higher sensitivity compared to western blot. These findings further support the advantages of using PICO for sensitive and accurate protein detection.
